Tangara callophrys is monotypic (Isler and Isler 1987, Dickinson and Christidis 2014).

Opal-crowned Tanager is a member of the speciose genus Tangara. This genus includes 49 species, more species than any other genus of exclusively Neotropical birds (Isler and Isler 1987). Isler and Isler (1987) subdivided Tangara into 13 species groupings based on geographic range, appearance, behaviors (with an emphasis on foraging behaviors), vocalizations, and nest sites. Opal-crowned tanager belongs to Isler and Isler’s species group 13, along with Tangara velia (Opal-rumped Tanager). These two species were formally classified together in a separate genus (Tangarella). Phylogenetic analysis of DNA sequence data confirms that these two species are sister taxa (Burns and Naoki 2004, Sedano and Burns 2010, Burns et al. 2014). In addition, these genetic studies showed that Tangara chilensis (Paradise Tanager), previously considered a member of species group 3, is closely related to the T. callophrys/T. velia clade.
